高脂饮食诱导的胰岛素抵抗对大鼠血浆同型半胱氨酸水平的影响

摘    要:目的:研究高脂饮食诱导的胰岛素抵抗(IR)对血浆同型半胱氨酸(Hcy)的影响。方法:将16只 Wistar大鼠随机分为2组:对照组给予普通饲料喂养;高脂组:喂高脂饲料诱导胰岛素抵抗动物模型。各组动物喂 食11周时,分别做空腹血糖、葡萄糖耐量实验后处死。测定血糖、胰岛素水平,用HOMA模型评价胰岛素抵抗指 数。同时喂食前和喂食11周后测定血浆Hcy水平。结果:高脂组体重变化、空腹胰岛素、空腹血糖、胰岛素抵抗指 数均有显著高于对照组(P<0.05)。两组间血浆Hcy也有显著差异,高脂组(35.7±14.1)μmol/L]高于对照组 (9.95±2.40)μmol/L]。相关分析显示空腹胰岛素、空腹血糖、胰岛素抵抗指数均与血浆Hcy水平显著相关 (P<0.05)。多元回归分析显示仅空腹血糖和胰岛素抵抗指数是血浆Hcy水平的独立相关因素。结论:高脂诱导 的胰岛素抵抗与大鼠血浆Hcy水平升高有关,并可能是胰岛素抵抗与心、脑血管疾病危险增加的原因之一。

Abstract:Objective:To determine whether plasma homocysteine (tHcy) levels were related to insulin resistance induced by high-fat diet in wistar rats. Methods:16 Wistar rats were randomized into 2 groups : control group(n=8) was fed with normal feeds, high-fat diet was given to high-fat group(n=8). 11weeks later, glucose tolerance test were performed, and serum insulin, fasting glucose and plasma homocysteine were detected. Viscus adipose was weighted and then calculated the ratio of viscus adipose and the body weight. Results:fasting glucose, fasting insulin, insulin resistance index (IRI), and the viscus adipose were significantly higher in high-fat group than that in control group(P<0.01). And the plasma homocysteine was significantly lower in control group than that in high-fat group. Correlation analysis showed that fasting glucose, fasting insulin, IRI, were all the factors influenced the plasma homocysteine. Stepwise regression test showed only fasting glucose(r=0.504,P=0.031) and IRI (r=0.302,P=0.046)independently affected the level of plasma homocysteine.Conclusion:We conclude that insulin resistance correlates significantly with plasma tHcy in high-fat feeding rats. This relationship may need to be considered when evaluating the role of plasma homocysteine as a risk factor in patients with type 2 diabetes.
