烟酸对大鼠脑缺血再灌性记忆再现障碍的保护作用

摘    要:用操作性条件反射法,观察烟酸对大鼠脑缺血再灌注引起的操作性记忆再现障碍的保护作用.用动脉夹将大鼠双侧颈总动脉夹闭20min后放开,造成脑缺血再灌注损伤.脑缺血对照组大鼠固定比率(FR4)操作反应率下降,操作潜伏期延长,辨别性操作正确率下降,7d后无明显恢复.脑缺血前后均给予烟酸10,20mg·kg-1·d-1各7d,大鼠的上述变化轻于脑缺血后给予烟酸10,20mg·kg-1·d-1共7d的大鼠.尤其高剂量组缺血再灌造成的损害轻,恢复快.提示烟酸对大鼠脑缺血再灌性记忆再现障碍有保护作用

Abstract:Protective effects of nicotinic acid on impairment of memory retrial induced by brief occlusion of bilateral common carotid artery for 20 min followed by reperfusion were observed using operative conditional reflex in rats. Wistar rats were trained to fulfil performance of fixed ratio(FR4) and discriminating operative activity before cerebral ischemia- reperfusion. Two groups of animal were given nicotinic acid 10 and 20 mg·kg-1·d-1 sc once a day from 7d before to 7d after cerebral ischemia respectively. Other two groups were administered by nicotinic acid 10 and 20 mg·kg-1·d-1 sc once a day for 7d after cerebral ischemia. It was found that the performance baseline was disturbanced, the performing latency was prolonged and the discriminating ability was impaired after cerebral ischemia-reperfusion. The performance disorders in cerebral ischemia-reperfusion rats were more severe than those of rats treated with nicotinic acid. Those operation memory dysfunctions could be restored gradually to normal level by nicotinic acid treatment during the following 7d observation. The protective effects of nicotinic acid on cerebral ischemia-reperfusion showed a dose-dependent manner. It is indicated that nicotinic acid improves the restore of disorders of memory retrial induced by cerebral ischemia-reperfusion in rats.
