温经通络法对CIA大鼠关节滑膜组织VEGF及其受体基因表达的影响

摘    要:目的观察温经通络法对CIA大鼠关节滑膜组织血管内皮生长因子(VEGF)mRNA及其受体VEGFR-1mRNA、VEGFR-2mRNA、VEGFR-3mRNA表达的影响。方法建立CIA大鼠模型,正常组、模型组各30只蒸馏水灌胃,对照组30只雷公藤多苷灌胃,大、中、小剂量组各30只分别用芍甘附子汤加味0.708、1.42、2.83g/kg体重灌胃,采用realtimePCR法检测大鼠膝关节滑膜组织VEGFmRNA及其受体VEGFR-1mRNA、VEGFR-2mRNA、VEGFR-3mRNA表达的相对含量。结果模型组VEGF、VEGFR-1、VEGFR-2、VEGFR-3表达水平与正常组比较均有明显增高,治疗大、中、小剂量组和对照组VEGF、VEGFR.1、VEGFR-2、VEGFR-3表达水平与模型组比较均有明显下降。结论温经通络法对CIA有明显的治疗作用,其机制可能与抑制VEGF及其受体表达有关。

Abstract:Objective To observe the effects of warming and activating meridian method on expression of vascular endothelial growth factor (VEGF) mRNA and its receptors VEGFR-1 mRNA, VEGFR-2 mRNA, VEGFR-3 mRNA in joint synovial tissue in CIA rats. Methods The CAI models of rats were established. With 30 rats in each group, intragastric administration of distilled water was applied in the normal group and model group; intragastric administration of tripterygium glycosides was adopted in the control group; intragastric administration of modified Shaogan Fuzi decoction was used in the high-dose group (2. 83 g/kg), medium-dose group (I. 42 g/kg) and low-dose group (0. 708 g/kg), respectively. The real-time PCR was employed to measure the relative content of expression of VEGF mRNA and its receptors VEGFR-1 mRNA, VEGFR-2 mRNA, VEGFR-3 mRNA in synovial tissue of knee. Results The expression of VEGF, VEGFR-1, VEGFR-2 and VEGFR-3 in the model group was significantly higher than that in the normal group. The expression of VEGF, VEGFR-1, VEGFR-2 and VEGFR-3 in the high-dose group, medium-dose group, low- dose group and control group was obviously lowered than that in the model group. Conclusion The warming and activating meridian method has obvious therapeutic effect on CIA, which could be related with inhibiting the expression of VEGF and its receptors.
