摘    要:目的 利用二维电泳-质谱技术分析Long Evans大鼠初级视皮层中与眼优势枉终止相关的蛋白质,探索蛋白质组学技术在视觉可塑性研究中的应用。方法取100d龄Long Evans大鼠通过向其初级视皮层中灌注硫酸软骨素酶建立成年眼优势柱移动的动物模型,然后分别提取22d龄大鼠、100d龄大鼠和100d龄硫酸软骨素酶灌注大鼠的初级视皮层灰质总蛋白,利用二维电泳-质谱技术分析与眼优势柱终止相关的蛋白质。结果 通过双向电泳确定了与眼优势柱终止相关的4个蛋白质点,质谱分析鉴定了其中2种蛋白质。这2种蛋白为磷脂酰乙醇胺结合蛋白和胶质纤维酸性蛋白。结论 二维电泳-质谱技术可以有效地分离和鉴定视皮层中的特殊蛋白质,随着技术的进步和完善,蛋白质组学技术必将在视觉可塑性研究中占有重要地位。

Abstract:Objective To analyze the protein related with ocular dominance plasticity termination in visual cortical area 17 of Long Evans rats by two-dimensional gel electrophoresis and mass spectrometry,and discuss the application of proteome technology in the study of visual plasticity.Methods Adult ocular dominance moving model was established by perfusing the chondroitinsulphate proteoglycan into the visual cortex of 100-day-old Long Evans rats,then the protein in the visual cortex was extracted from 22-day-old rats,100-day-old rats and 100-day-old perfused rats respectively,and the protein related with ocular dominance plasticity termination was analyzed by two-dimensional gel electrophoresis and mass spectrometry.Results Four protein spots that related with ocular dominance plasticity termination were found by gel-image analysis,two proteins were identified by mass spectrometry,one was phosphatidylethanolamine binding protein,the other was Glial fibrillary acidic protein.Conclusion Two-dimensional gel electrophoresis and mass spectrometry is effective to separate and identify the special visual cortex proteins.With the progress of proteomics technique,it will be very important in the study of visual plasticity.
