乙酰化转移酶抑制剂对人肝癌细胞SMMC-7721基因表达的影响

摘    要:目的通过研究乙酰化转移酶抑制剂(TSA)对人肝癌细胞SMMC-7721基因表达谱的影响,初步探讨TSA对肝癌细胞抑制作用的机制。方法用含有3072个样点,覆盖2748个肿瘤相关的基因的XproTMHC-Ⅲplus基因表达谱芯片来研究TSA对肝癌细胞基因表达的影响。分离纯化采作任何处理的肝癌细胞和用TSA处理过的肝癌细胞的mRNA。制备荧光标记的cRNA靶物(Cy3或Cy5标记)。靶物与基因芯片杂交。用Axon4000B型扫描仪进行芯片荧光信号扫描。利用计算机分析用TSA处理过的肝癌细胞和未处理的肝癌细胞基因表达的差异。结果在2748个基因中,有显著性差异的基因共191个,其中TSA处理组相对于未处理组基因上调2倍以上的有173个,TSA处理组相对于未处理组基因下调0.5倍以下的基因18个。结论TSA通过调控多个基因共同作用采抑制肝癌细胞的生长。这些基因在肝癌的发病中可能起着非常重要的作用。

Abstract:Objective] To study the machenism of inhibition of histone deacetylase inhibitors (TSA) to hepatoma using microarry. Methods] The total RNA of the hepatoma cells treated with TSA or untreated were abstracted. Analyzed the gene profile changes with XproTMHC-III plus gene chips kits. Results] Among 2748 target genes, there were 191 genes whose expression were different between treated group and untreated group, 173 of 191 were 2 times upregulated, 18 of 191 were 0.5 times down regulated in the treatment group compared to the untreated group. Conclusions] TSA inhibited the growth and induced the cell death of hepatoma cells through multiple genes, all these genes work together to produce the results. They are all important in the carcinogenesis of hepatoma.
