小鼠胸腺树突状细胞系MTSC4诱导早期T细胞株C320表达IL-2R

摘    要:通过流式细胞仪(FACS)分析了我室自建的小鼠胸腺树突状细胞系MTSC4与小鼠肿瘤性早期T细胞株C320细胞共育后,C320细胞表达IL-2R的状态,分析了ConA和PHA对C320及与MTSC4共育后的C320表达IL-2R的诱导促进作用以及IL-2R+C320细胞在脱离外源性刺激后其比例的变化。MTSC4及ConA和PHA在不同程度上抑制C320细胞的无限制增殖,ConA和PHA对与MTSC4共育后的C320细胞的抑制增殖作用更强。

Abstract:The CD25 (IL-2R ,P55)-pgp-1 CD4-CD8- CD3 LFA-1 mouse Pre-T lymphocyte clone C320 could be in-duced to express IL-2R by co-culture with mouse thymic dendritic cell line MTSC4. The fraction of C320 cell re-covered after 7 days and 14 days from these cultures could express IL-2R analysed by FACS. The observation indi-cated that these in vitro gencrated IL-2R C320 cells were responsive to Con A and PHA to express more IL-2R.Cell surface phenotyping suggests that IL-2R pre-T cells are more mature than IL-2R-pte-T cell. The rate ofIL-2R cell was reduced without any exogenous stimulants.MTSC4,Con A and PHA could inhibit unlimited pro-liferation of C320 calls in different degrees.
