PTEN体外转染对乳腺癌细胞ZR-75-1细胞周期和增殖的影响

摘    要:目的探讨外源野生型PTEN基因对人乳腺癌细胞ZR-75-1细胞周期和增殖的影响,阐明PTEN基因抑制肿瘤细胞增殖的机制。方法利用脂质体介导法将重组质粒pBP-wt-PTEN导入人乳腺癌细胞ZR-75-1中,嘌呤霉素筛选稳定表达细胞株并扩增培养;采用细胞计数法检测肿瘤细胞增殖速度;流式细胞术检测细胞周期的变化。结果经嘌呤霉素筛选得到稳定表达细胞株;重组质粒pBP-wt-PTEN稳定转染可明显抑制ZR-75-1细胞的体外增殖,细胞周期明显改变,细胞从G1期到S期发生抑制,并在G1期峰前出现1个较为明显的凋亡峰。结论转染外源性野生型PTEN基因可使ZR-75-1细胞周期阻滞于G1期,并可抑制肿瘤细胞增殖。

Abstract:Objective To investigate the effect of exogenous wild type PTEN(phophatase and tensin homolog deleted on chromosome) gene transfer on the cell cycle progression and proliferation of ZR-75-1 human breast cancer cells,and to illustrate the mechanism that PTEN gene inhibits the proliferation of tumor cells.Methods pBP-wt-PTEN recombinant plasmids were transfected into ZR-75-1 cells by Lipofectamine.The stable express cells were selected by puromycin.The proliferation of tumor cells were examined by cell counted while the changes of cell cycle progression were by flow cytometry.Results Stable express cells were obtained by puromycin selection.The growth of ZR-75-1 was inhibited significantly by pBP-wt-PTEN stable transfering in vitro.The progression of cell cycle was arrested from G_1 to S phase,and one visible apoptotic peak in front of the peak of G_1 phase was also found by FCM.Conclusion The growth of ZR-75-1 breast cancer cells can be arrested at G_1 phase,and it can be significantly suppressed by exogenous PTEN gene transfering.
