摘    要:目的探究人参皂甙Rg3对胰腺癌血管生成拟态作用。方法体外用鼠尾I型胶原蛋白建立三维培养体系,观察胰腺癌SW-1990、Panc-1、Bxpc-3、MiaPaCa-2四株细胞株形成血管生成拟态能力,应用PASR染色筛选出能够形成血管生成拟态的细胞株;用该株细胞建立体外模型,不同浓度人参皂甙Rg3(0、25、50、100、200μmol/L)处理模型,观察人参皂甙Rg3对该细胞株形成血管生成拟态的影响;进一步利用荧光定量PCR技术和Western blotting检测血管生成拟态相关指标MMP-2、MMP-9表达。结果胰腺癌SW-1990、Panc-1、Bxpc-3、MiaPaCa-2四株细胞株中,SW-1990在体外用鼠尾Ⅰ型胶原蛋白建立三维培养体系中培养72h后能够形成血管生成拟态。荧光PCR技术和Western blotting检测发现,与对照组相比,用药组MMP-2和MMP-9在mRNA水平和蛋白水平表达均下降:与空白组相比,各用药组MMP-2 mRNA水平和蛋白水平表达差别均有统计学意义(P〈0.05);MMP-9在mRNA水平和蛋白水平表达情况,25μmol/L组与空白组比较无统计学意义(P〉0.05),50μmol/L组、100μmol/L组、200μmol/L组与空白组比较差别有统计学意义(P〈0.05)。结论人参皂甙Rg3能够抑制胰腺癌血管生成拟态的形成,下调MMP-2和MMP-9的表达可能是其机制之一。

Abstract:Objective To explore the effect of Ginsenoside Rg3 on pancreatic cancer vasculogenic mimicry. Methods In vitro rat tail collagen type I was used to build three-dimensional culture system, SW-1990, Pane-1, Bxpc-3, MiaPaCa-2 cells were seeded onto three-dimensional culture system. PAS staining was conducted to observe the ability of cells form vasculogenic mimicry and screen out which cell can form vasculogenic mimicry. Vitro models were established by this cell lines. In order to observe the effects of ginsenoside Rg3 on vascular mimicry effects, vitro models were treated with various concentrations of ginsenoside Rg3 (0, 25, 50, 100, 200 μmol/L). FQ-PCR was used to observe the mRNA expression of MMP-2, MMP-9. Western blotting was used to observe the protein expression of MMP-2, MMP-9. Results In pancreatic cancer cell lines SW-1990, Panc-1, Bxpc-3 and MiaPaCa-2, only SW-1990 could form vasculogenic mimicry. Compared with the control group, FQ- PCR and Western blotting results showed that MMP-2 mRNA and protein expressions in drug groups were decreased significantly (P〈0.05); MMP-9 mRNA and protein expressions in drug groups were decreased to a different extent, except for 25 μmol/L group, 50, 100 and 200 tarnol/L group were decreased significantly (P 〈0.05). Conclusion Ginsenoside Rg3 has the ability to inhibit the growth of pancreatic cancer vasculogenic mimicry by regulating the vasculogenic factors, which can be related to the down regulation of MMP9 and MMP2 expression.
