摘    要:目的 建立猪微卫星标记的研究方法 ,分析微卫星标记的多态性特点 ,为基因连锁分析和数量性状位点(QTL)定位提供依据。方法 采集大白×梅山猪资源家系 10头F0 代、 2 8头F1代及 14 7头F2 代猪的前腔静脉血 10ml,苯酚氯仿法提取DNA。用分布 6条染色体的 4 8对微卫星引物 ,对上述样品DNA进行PCR扩增。扩增产物经聚丙烯酰胺凝胶电泳后银染显色 ,干胶保存、基因型判定。结果 优化了 4 8个微卫星标记PCR反应条件。微卫星标记平均等位基因数 3 6 3个 ;多个等位基因片段大小超过USDA -MARC在网上公布的范围 ;平均有信息减数分裂数为 2 4 7;平均微卫星多态信息含量 0 5 4 6 9,考虑同一连锁群所有标记时 ,平均多态信息含量为 0 7370。结论 微卫星标记具有很高的信息含量 ,将对后续的遗传图谱构建及QTL区间作图起到很好的支持作用。

Abstract:Objective In order to establish the method for microsatellite markers in pigs and provide a basis for microsatellite markers applied in genetic map and QTL(quantitative trait loci)ma pping. Methods Sample DNA of 10 F 0,28 F 1 and 147 F 2 offspring in a resource family crossed by Large White and Meishan pigs were extracted from blood.In this paper,48 pairs of microsatellite primers in pig covering 6 chromosomes were used to PCR amplification and genotyped. Results The PCR conditions for 48 microsatellite markers in the resource family were selected.Average allele number was 3 63,average informative meiosis 247,average polymorphic information content 0 5469,and 0 7370 using all information in one linkage.Many allele sizes exceeded the range published by USDA-MARC. Conclusions The characterization of these microsatellite markers was shown that they were highly polymorphic and these markers could be used to construction of linkage map and detection of quantitative trait loci(QTL).
