胃癌中DAPK1表达及其与临床病理因素的相关性研究

目的 检测胃癌中死亡相关蛋白激酶1(DAPK1)基因表达水平,并研究其与临床病理因素间的相关性.方法 采用免疫组织化学法(SP)检测胃癌肿瘤组织和癌旁正常组织中DAPK1基因表达,采用半定量评分法评估,分析DAPK1表达水平与胃癌临床病理因素间的相关性.结果 胃癌肿瘤组织标本中,DAPK1蛋白质低、中和高表达分别有52、19、5例,评分为(3.7±1.3)分,癌旁正常组织标本中,低、中表达和高表达分别为4、15、57例,评分为(9.4±2.1)分,胃癌肿瘤组织和癌旁正常组织评分差异具有统计学意义(P<0.01).胃癌肿瘤组织中DAPK1表达在性别、年龄和不同肿瘤部位间差异无统计学意义(P>0.05),而组织学分级、TNM分期、肿瘤最大径、淋巴结转移及远处转移对DAPK1表达的影响具有统计学意义(P<0.05);胃癌肿瘤组织DAPK1表达评分与组织学分级、TNM分期、肿瘤最大径、淋巴结转移和远处转移呈负相关(P<0.05).结论 DAPK1在胃癌中的表达与肿瘤细胞的生长、浸润及转移紧密相关,可作为胃癌诊断、病情及预后评估的指标.

Expression level of DAPK1 in gastric cancer and its correlation with clinicopathologic factors

Objective To study the expression level of death-associated protein kinase 1(DAPK1) in gastric cancer and its correlation with clinicopathologic factors.Methods Expression level of DAPK1 was detected with immunohistochemistry(SP) in gastric cancer tissues and normal tissues adjacent to carcinoma.The correlation between its expression level in gastric cancer and clinicopathologic factors was analyzed.Results The low,median and high expression of DAPK1 in gastric cancer were 52,19 and 5 cases respectively,with expression score of 3.7±1.3,while there were 4,15 and 57 cases in normal tissues adjacent carcinoma with expression score of 9.4±2.1.The differences of DAPK1 expression between different gender,age and tumor site were not statistically significant(P>0.05),while between different histology grade,TNM staging,tumor size,lymphatic metastasis and distant metastasis were statistically significant(P<0.05).DAPK1’s expression was correlated negatively with histology grade,TNM staging,tumor size,lymphatic metastasis and distant metastasis(P<0.05).Conclusion Expression of DAPK1 in gatric cancer tissues might be correlated with the growth,infiltration and metastasis of cancer,which could act as a marker for the diagnosis,condition and prognosis evaluation of gastric cancer.
