化浊祛瘀汤治疗酒精性肝病的实验研究

摘    要:目的探讨化浊祛瘀汤对酒精性肝病大鼠影响的机理。方法通过乙醇梯度灌胃的方法建立酒精性肝病(ALD)大鼠模型,并使用化浊祛瘀汤给大鼠灌胃,观察化浊祛瘀汤对酒精性肝病肝损伤大鼠NF—κB表达,NF—α、CAT等水平的影响,并进行HE染色,观察肝脏组织基本病理改变。结果模型组大鼠NF—κB表达明显增强(P〈0.01),TNF—α含量显著增加(P〈0.01),CAT含量明显降低(P〈0.01);化浊祛瘀汤治疗组NF—κB表达明显减弱,TNF—α含量明显降低(P〈0.01),CAT含量明显升高(P〈0.01);中药高剂量组与西药组比较有显著性差异(P〈0.01)。结论化浊祛瘀方药能够抑制酒精性肝病时NF—κB、TNF—α的表达,改善ALD脂质过氧化过程。

Abstract:Objective To explore the mechanism of Huazhuo Quyu Tang on alcoholic liver disease (ALD) in rats. Methods Alcohol - gradient gastric perfusion was used to establish ALD rat model. Huazhuo Quyu Tang was administered by gastric perfusion in rats to observe the impacts of the decoction on NF -κB expression and the levels of TNF - α and CAT in ALD rats. By HE staining, the basic pathological changes of liver tissue was observed. Results In model group, NF -κB expression increased apparently ( P 〈0.01) , TNF -α level increased apparently (P 〈 0.01 ) and CAT reduced apparently (P 〈 0.01). In Huazhuo Quyu Tang treatment group, NF - κB expression decreased apparently( P 〈0.01) , TNF - α level reduced apparently( P 〈 0.01 )and CAT increased apparently( P 〈 0.01 ). The significant difference presented in comparison between high-dose Chinese herb group and western drug group (P 〈 0.01 ). Conclusions Huazhuo Quyu Tang can inhibit the expressions of NF - κB and TNF -α and improve lipid peroxidation of ALD.
