龙胆苦苷血药浓度测定法在磷脂复合物制备工艺研究中的应用

目的 建立大鼠血浆中龙胆苦苷(gentiopicroside,GTP)浓度的RP-HPLC测定方法并将其用于龙胆苦苷磷脂复合物制备工艺研究.方法 建立大鼠血浆中龙胆苦苷浓度的测定方法并对其进行评价;制备龙胆苦苷磷脂复合物,并将其溶解于中链椰油甘油酯(Imwitor 742),制得磷脂复合物油溶液;分别进行灌胃给予三种不同药脂比龙胆苦苷磷脂复合物油溶液的大鼠体内相对生物利用度评价,筛选最优药脂比.结果 所建立的大鼠血浆中龙胆苦苷浓度的测定方法专一性、准确度、精密度、稳定性等均符合生物样品测定要求;以大鼠生物利用度为评价指标,可以反映出不同药脂比制备的磷脂复合物之间存在的质量差异.结论 所建立的血浆药物浓度测定方法可用于龙胆苦苷新制剂的制备工艺研究和质量评价.

Analysis method of gentiopicroside in rat plasma and its application in investigation of preparation technology of phytosome
