1,25-二羟胆钙化醇与其受体基因表达关系的研究

摘    要:目的:研究DHCC与其受体基因表达的关系。方法:幼年大鼠经腹腔注射DHCC后,在3、6、12和24h处死,并分别提取小肠、肾脏、心脏、肝脏和脾脏等总RNA,用生物素标记的DHCCR基因为探针进行Dotblot杂交与用32P标记的DHCCR基因为探针进行Northernblot杂交检测DHCCRmRNA水平。结果:给药后第6h靶器官中的DHCCRmRNA水平明显增高。检测DHC-CR分布时发现小肠DHCCRmRNA含量最高,肾脏其次,心脏为第三位,肝脏与脾脏中未见表达。结论:DHCCR基因表达不仅出现在像小肠、肾脏等传统的靶器官,而非靶器官的心脏也有DHCCRmRNA信号。提示心脏可能是一个待定的DHCC靶器官。

Study on relationship between 1,25-dihydroxycholecalciferol and its receptor gene expression
Abstract:Objective:The relationship between 1,25 dihydroxycholecalciferol (DHCC) and its receptor (DHCCR) gene expression was investigated.Methods:Infant rats were injected with DHCC.Total RNA of intestine,kidney,heart,liver,and spleen in rat was extracted at 3,6,12 and 24 h after administration.Dot blot small hybridization with biotin labelled DHCCR gene probe and Northern bolt hybridization with 32p labelled DHCCR gene probe were used to determine mRNA levels.Results:DHCCR mRNA level increased from 6 h after DHCC was given.For the distribution of DHCCR,it was found that DHCCR mRNA level in small intestine was the highest,the level in kidney came the second,in heart it was the third.In liver and spleen DHCCR gene did not express.Conclusion:Based on these findings,the DHCCR gene expressed not only in the traditional target organs,such as small intestine and kidney,but also in heart which used to be considered as non target organ of DHCC. Heart possibly is a putative target organ of DHCC.
