Sanger测序法分析兰州地区112例丙型肝炎患者基因亚型

摘    要:目的探讨兰州地区HCV感染者基因亚型分布及其临床价值。方法选取了112例HCV感染者血清标本,测定HCV RNA病毒载量、Sanger测序法检测HCV基因亚型分布、ALT、AST等肝功能相关指标。结果 112例标本中血清学分型率为80. 4%,其中1b型占39. 3%,2a型占35. 7%,3a型占3. 6%,6a型占0. 9%,6g型占0. 9%,未检出组占19. 6%。基因1b型、2a型、3a型与未检测组HCV RNA病毒载量比较,差异有统计学意义(P <0. 05);其中1b型、2a型和3a型与未检出组ALT水平比较,差异有统计学意义(P <0. 05)。结论兰州地区HCV感染以1b型为主,其次为2a型、3a型、6a型、6g型。且HCV基因亚型与ALT、AST等肝功能指标及HCV RNA病毒载量具有一定的相关性,为该地区丙型肝炎的预防和治疗提供部分理论依据。

Abstract:Objective To investigate the distribution and clinical value of gene subtypes in patients with hepatitis C( HCV) in Lanzhou area. Methods The serum samples of 112 patients with HCV were selected as the study objects. HCVRNA viral load was measured. The HCV gene subtype distribution and liver function related indexes of ALT,AST were detected by Sanger sequencing. Results The serological typing rate of 112 samples was 80. 4%,of which 1b type accounted for 39. 3%,2a type accounted for 35. 7%,3a type accounted for 3. 6%,6a type accounted for 0. 9%,6 g type accounted for 0. 9%,and undetected group accounted for 19. 6%. There was statistical significance on the difference in the HCV RNA viral load between the gene type 1 b,2a,and 3a and the undetected group( P < 0. 05). Among them,the ALT levels of type 1b,2a,and 3a were compared with the undetected group,and the differences were statistically significant( P < 0. 05). Conclusion In Lanzhou area,1b was the main type of HCV infection,followed by 2a,3a,6a,6g. The subtype of HCV gene was correlated with the liver function index such as ALT,AST and the load of HCV RNA virus,which can provide some theoretical basis for the prevention and treatment of hepatitis C in this area.
