miR-34a和miR-21在宫颈癌中的表达及其临床意义

目的探讨微小核糖核酸-34a(miR-34a)和微小核糖核酸-21(miR-21)在宫颈癌(CC)患者中的表达及其临床意义。方法采用实时荧光定量PCR检测受试者外周血浆中的miR-34a和miR-21表达水平。使用SPSS 17.0软件进行t检验,并使用受试者工作特征曲线(ROC)评估诊断效能。结果CC组m iR-21表达水平显著高于对照组(P<0.01),miR-34a表达水平显著低于对照组(P<0.05):miR-21、miR-34a的表达与临床分期和淋巴结转移相关(P<0.01);miR-21和miR-34a的曲线下面积(AUC)分别为0.881和0.911,两者均具有较高的诊断效能。结论miR-21可能为促癌基因,miR-34a为抑癌基因,miR-21和miR-34a参与了宫颈癌的发生发展,它们对宫颈癌的诊断和监测具有重要的临床价值。

Expression and clinical significance of miR-34a and miR-21 in cervical cancer

Objective To investigate the expression and clinical significance of microRNA-34a(miR-34a)and microRNA-21(miR-21)in cervical cancer(CC)patients.Methods The expression levels of miR-21 and miR-34a in peripheral plasma were measured by qRT-PCR.The data were statistically analyzed using SPSS 17.0 software.The two groups of measurement data were compared by t test,and the diagnostic performance was evaluated by the receiver operating curve(ROC).Results The expression level of miR-21 in CC group was significantly higher than that in the control group(P<0.01)and the expression level of miR-34a was significantly lower than that in the control group(P<0.05).The expressions of miR-21 and miR-34a were correlated with clinical stage and lymph node metastasis(P<0.01).The area under the curve(AUC)of miR-21 was 0.881,and the AUC of miR-34a was 0.911,both of which had higher diagnostic potency.Conclusion MiR-21 may be a tumor-promoting gene,miR-34a is a tumor suppressor gene,miR-21 and miR-34a are involved in the development of cervical cancer,and have important clinical value in the diagnosis and monitoring of cervical cancer.
