先天和后天感染鸭乙肝病毒的广州麻鸭外周血中病毒血症的动态比较及应用

摘    要:为寻求较理想的动物乙型肝炎模型,在研制成功广州麻鸭乙型肝炎模型基础上,比较先天和后天感染鸭乙肝病毒的广州麻鸭外周血中病毒血症的差异。方法是采用斑点杂交方法(dotblot)检测了广州麻鸭先天和后天感染鸭乙型肝炎病毒(DHBV)后外周血中DHBV-DNA的动态。结果:先天感染的广州麻鸭外周血中病毒血症水平较高,出生后(即感染后)26d时DHBV-DNA下降到一较低的水平,72d时仍维持较低水平。出生后1d感染DHBV的广州麻鸭,感染后9d时血中开始出现低水平的DHBV-DNA,11d时达到高峰,之后逐渐下降,26d时下降到一较低的水平,到72d时仍维持低DHBV-DNA水平。并采用后天感染DHBV的广州麻鸭作为鸭乙肝模型筛选抗乙肝中草药,以无环鸟苷为阳性对照药,蒸馏水为阴性对照药,发现每次阳性对照组和阴性对照组均成立,证明该模型重复性良好。并发现台湾单纯叶下珠、虫草和树舌均有降低DHBV-DNA的作用。

Abstract:DHBV-DNA in peripheral blood of Guangzhou brown spot ducks with congenital and acquired infection of duck hepatitis B virus was observed dynamically by dot blot method.The results showed that the level of DHBV-DNA was higher in the congenitally infected ducks and decreased to a lower level on the 26 th day up to the 72 nd day.In the one-day-old duckling infected with positive DHBV serum of Shanghai brown spot ducks, the level of DHBV-DNA became lower on the 9 th day, then rose to the peak on the 11 th day,and decreased to a lower level on the 26 th day up to the 72 nd day.We also used the one_day_old postnatally infected duckling as the model of HBV infection for anti_HBV drug screening test and found that three Chinese herbal medicines,Taiwan's Phyllanthus simplex,Paecilomyces sinensis Chen.Xiao et Shi var.grossotilbi Chen and Ganoderma applanatum(Pers.ex Tray)Pat,possessed an inhibitory effect on DHBV_DNA.
