胃癌组织中Toll样受体4/9的表达与肿瘤病理分期和转移的相关性

摘    要:目的:探讨Toll样受体(Toll-like receptors,TLR)4和TLR9在胃癌组织中的表达及其与临床病理特征的关系。方法:收集2009年1月至2009年10月在解放军第174医院手术切除的42例胃癌及其癌旁(距癌灶边缘5 cm)组织标本。RT-PCR和Western blotting检测胃癌组织中TLR4、TLR9在mRNA和蛋白水平的表达。结果:胃癌组织中TLR4和TLR9 mRNA表达高于癌旁组织(1.29±0.03 vs 0.53±0.02,0.99±0.04 vs 0.22±0.05;均P<0.05),Ⅲ期+Ⅳ期胃癌组织中TLR4和TLR9mRNA表达显著高于Ⅰ期+Ⅱ期(P<0.05),有远处转移的胃癌组织中TLR4和TLR9 mRNA表达显著高于无远处转移者(P<0.05);TLR4和TLR9 mRNA的表达水平与患者的年龄、性别、淋巴结转移没有相关性。胃癌组织中TLR4和TLR9蛋白的表达高于癌旁组织(1.36±0.05 vs 0.48±0.04,1.12±0.05 vs 0.34±0.03;均P<0.05),Ⅲ期+Ⅳ期胃癌组织TLR4和TLR9蛋白的表达显著高于Ⅰ期+Ⅱ期(P<0.05)。结论:TLR4和TLR9在胃癌组织中高表达,其表达与胃癌分期、远处转移具有相关性,表明TLR4和TLR9可能参与了胃癌的发生和发展,可能为胃癌的治疗提供一种潜在的靶点。
