腰神经根受压后花生凝集素结合分子在大鼠肌肉神经再生期的分布变化

摘    要:目的 研究腰神经根损伤后 ,花生凝集素结合分子 (penutagglutinin bindingmolecules ,PNA BMs)在大鼠比目鱼肌神经肌肉接头部的分布变化。方法 采用组织化学方法和激光共聚焦扫描显微镜技术及能与细胞外基质中配糖体 (glycoconjugate)特异性结合的PNA染色 ,在腰神经根受压后 10 ,2 0 ,30 ,及 60天 ,观察PNA BMs在神经肌肉接头部的分布 ,运用α bungarotoxin(α BTX)荧光结合剂显示运动终板 ,NIH图象分析技术测定PNA BMs与运动终板重叠面积。结果 肌肉失神经支配后 10天 ,神经肌肉接头部PNA染色大幅度消褪或消失 ,乙酰胆碱斑呈卷曲 ,不规则 ,或崩解。随着神经再支配的进行 ,乙酰胆碱斑形态逐渐趋于正常 ,PNA染色强度及范围也逐渐回升 ,最终局限于运动终板区域内。结论 PNA BMs在神经肌肉接头部的分布变化与肌肉失神经支配及神经再支配密切关联。

Abstract:Objective To study the changes in the distribution of Peanut Agglutinin Binding Molecules (PNA?BMs)in neuromuscular junction of soleus muscle after compression of the L 5 nerve root in rats. Methods Using histochemistry,confocal laser scanning techniques and the PNA staining that is known to specifically bind to some glycoconjugates in the extracellular matrix. The changes has been observed in a neuromuscular junction at 10d?20d?30d?and 60d after the L 5 nerve root was compressed. Motor endplates were visualized with fluorescein-conjuated a-bungarotoxin (α?BTX).The overlap area of PNA-BN s and measured with NIH image technique.Results 10 d after L 5 nerve root injury resulted in great reduce or disappearance of PNA staining and acetylchline receptor(AChR) plaques apparent shrinkage, disorgainize and fragmentation. reinnervation resulted in an AChR plaques display transitino to normality in size and shape. The range and intensity of PNA?BMs increased step by step.Conclusion The changes of PNA?BMs is relation to degeneration and regeneration in neuromuscular junctions.
