加味越婢加半夏汤对COPD模型大鼠血清IL-8和TNF-α的影响

【目的】研究加味越婢加半夏汤对慢性阻塞性肺病(chronic obstructive pulmonary disease,COPD)大鼠模型肺组织形态学以及血清白介素-8(IL-8)、肿瘤坏死因子-α(TNF-α)的影响,为加味越婢加半夏汤的临床应用提供实验依据。【方法】将40只Wistar大鼠随机分成模型组、强的松组、加味越婢加半夏汤低剂量组、加味越婢加半夏汤高剂量组,每组10只,以烟熏及气管内滴注脂多糖的方式建立大鼠COPD模型,HE染色对各组大鼠进行肺部组织学评分,ELISA法检测大鼠血清中IL-8、TNF-α的含量。【结果】模型组肺部炎性细胞浸润,肺泡扩大融合,数量减少;血清中IL-8和TNF-α高表达,各治疗组以上改变均明显减轻,以强的松组和加味越婢加半夏汤高剂量组最为显著(P<0.01),且二者治疗效果无明显差异。【结论】加味越婢加半夏汤能有效缓解COPD大鼠模型肺部炎症,降低了COPD大鼠体内细胞因子IL-8和TNF-α水平,上述作用具有一定的量效关系,即随给药剂量的增加有加强趋势。

Influence of Supplemented Spleen-effusing Decoction plus Pinellia on pathomorphological changes and contents of cytokines in rats with COPD

【Objective】To investigate the influence of supplemented spleen-effusing decoction plus pinellia on pathomorphological changes and the contents of cytokines（IL-8,TNF-α）in rats with COPD and provide experimental basis for clinical application.【Methods】40 Wistar rats were divided randomly into 4 groups：model control group,prednisone group,supplemented spleen-effusing decoction plus pinellia low dose group and supplemented spleen-effusing decoction plus pinellia high dose group,each group with 10 rats.Then we copied COPD rats model by being smoked and inhaled with LPS.At the same time,each group was given relevant medicines.After four weeks,we killed all the rats.And then we surveyed the contents of cytokines（IL-8,TNF-α）in serum.【Results】Model control group had unbalance in the contents of cytokines in serum.Compared comprehensively,prednisone group were better than other groups,supplemented spleen-effusing decoction plus pinellia high dose group secondly,supplemented spleen-effusing decoction plus pinellia low dose group worst.【Conclusion】Supplemented spleen-effusing decoction plus pinellia could alleviate the airway inflammation and affected the contents of cytokines in serum.Above-mentioned action can be reinforced by increased dose.
