摘    要: 目的 对不同接种方法 构建的小鼠L1210白血病模型的某些生物学特性进行比较研究,选择基本符合L1210细胞生物学特性,且一致性更好的接种方法.方法 常规培养小鼠急性淋巴细胞白血病细胞系L1210,分别给DBA/2 小鼠以静脉(A组)、皮下(B组)及腹腔注射(C组)3种途径接种,观察小鼠成瘤情况,定期计数各组小鼠外周白细胞数量并观察细胞形态改变,取濒死小鼠的肝、脾、肺、肾等脏器称重并制作病理切片.结果 各组小鼠成瘤率均为100%,A、B、C 3组成瘤天数分别为:14、7和14 d;平均存活天数分别为:(253±310)、(256±192)、(181±187) d.接种后2周外周血白细胞数目分别增加至(1832±201)、(2220±388)、(2796±713)×109/L,接种后3周外周血白细胞数目分别增加(2201±1196)×109/L (A组)和(7023±304)×109/L(B组);外周血涂片中均可见白血病细胞;各组濒死小鼠肝、脾有弥漫性白血病细胞浸润,正常组织结构破坏;肺和肾内有少量白血病细胞浸润,正常组织结构破坏不明显,肺脏有明显出血.结论 静脉、皮下、腹腔注射体外培养的L1210细胞于DBA/2小鼠均可成瘤.皮下注射成瘤时间短,存活时间长,可见明显的瘤结节;腹腔注射成瘤时间长,存活时间短,与L1210细胞传代的接种方式一致;静脉注射成瘤时间长,存活时间也长,与白血病血行播散的方式一致.
