隧道法:一种评价大鼠脑缺血模型神经功能的方法

摘    要:目的 本研究旨在评价一种新的、简便的、比较精细的大鼠脑缺血模型的行为学评分方法--隧道法.方法 用改良的大脑中动脉结扎法制作大鼠大脑中动脉缺血模型.TTC染色验证模型制作的有效性,缺血后2d、1周、2周、4周,分别用5分法和隧道法评价大鼠的神经功能,并测量大鼠的体感诱发电位和运动诱发电位.结果 TTC染色显示大脑中动脉分布区皮层梗死灶.5分法只能反映是否存在皮层损伤,但不能反映脑缺血后自发恢复的细微神经功能变化.隧道法双前肢和双后肢评分之和的峰值出现在缺血后2d和1周,1周后下降;双前肢和双后肢评分之差的高峰出现在缺血后1周,1周后下降.体感诱发电位显示术后缺血侧肢体的波形消失,缺血2周后有波幅恢复的趋势,潜伏期大于对照组;而运动诱发电位却显示反常的现象,即缺血侧的波幅反而高于未缺血侧,而潜伏期没有明显差别.结论 隧道法是一种比较简便、精确的评价大鼠脑缺血神经功能的方法,可以用于大鼠脑缺血模型的实验研究.

Abstract:Objective To evaluate a novel,convenient and sensitive gauging system suitable to cerebral ischemia animal model--trap channel test.Method The cerebral ischemia models were made by ligating the middle cerebral artery(MCA)of Sprague-Dawley rats.The ischemic injury was confirmed by TTC staining.The neurological functions of the ischemic rats were evaluated with 5-score system,trap channel test and somatosensory evoked potential(SEP)and motor evoked potential(MEP)on the 2nd day,1st week,2nd week and 4th week after surgery.Results TTC staining demonstrated an infarctive area in the cortex of MCA distribution.5-score system could confirm the cortex injury,but could not be used to distinguish the subtle recoveries of the neurological functions.In the trap channel test,the Sum of bilateral extremities scores showed a peak on the 2nd day and 1st week after surgery,and decreased after the 1st week;the difference of the scores between injured and intact extremities showed a peak on the 1st week after surgery,and decreased after the 1st week.All components of SEP disappeared within the first week after the surgery,while manifested a recovery trend after 2w post-surgery,however,the MEP showed a paradox,i.e.the amplitude of MEP of the injured lateral was higher than the counter-part of intact lateral,and there was no significant differentiation between bilateral MEP latency.Conclusions Trap channel test is a convenient and sensitive gauging system to assess the neurological functions of the ischemic rat models,and can be beneficial for the experimental studies of cerebral ischemia.
