正丁烯呋内酯PVA凝胶对豚鼠平喘作用的研究

摘    要:目的探讨正丁烯呋内酯制成聚乙烯醇(PVA)凝胶贴片透皮给药对豚鼠的平喘作用.方法精密量取一定量正丁烯呋内酯加入到由适量15%PVA、5%干淀粉、10%甘油组成的基质中,浇注于预制的模具中,得厚1.5 mm、直径15 mm每片含药量为20 mg的凝胶贴片.取哮喘预选试验合格的豚鼠随机分为空白贴片组、正丁烯呋内酯PVA凝胶贴片组和氨茶碱贴片组3组.各组将豚鼠背部脊柱两侧第3椎对称部位中心的皮肤上的毛剪去.取相应的贴片敷于上述暴露的皮肤表面,每天贴敷给药1次,每次8 h,连续给药5 d,末次给药2h后,测定各组哮喘潜伏期、跌倒休克期和病死率.结果正丁烯呋内酯聚乙烯醇凝胶贴膏透皮给药能延长豚鼠的哮喘潜伏期和跌倒休克期(P<0.01).结论正丁烯呋内酯透皮给药方便、有效.

Abstract:Objective:To study the antiasthmatic effect of n-butylidene phthalide for transdermal administration in guinea pigs.Methods:The gelatin paster contained n-butylidene phthalide(25 mg/tablet)was made from 15% of PVA,5% dry starch and 10% glycerin.Eighteen guinea pigs were randomly divided into three groups,blank paster group,n-butylidene phthalide PVA paster group and aminofilina paster group.Remove the hairs from the skin on the bilateral back in all of guinea pig groups.Put the paster on both sides of exposed skin 8 h/d for 5 days.After 2 h of final administration on the fifth day,the latency of induced asthma,shock stage and fatality rate were measured for each group.Results:The experimental data indicated that the n-butylidene phthalide gelatin paster could significantly prolong the latency of induced asthma and shock stage in guinea pigs(P < 0.01).Conclusion:The transdermal administration of n-butylidene phthalide gelatin paster is convenience and efficiency.
