琼枝麒麟菜多糖对辐射损伤小鼠细胞因子分泌的影响

摘    要:目的观察琼枝麒麟菜多糖(EGP)对辐射损伤小鼠细胞因子分泌的影响,为辐射损伤的治疗提供依据。方法将50只小鼠随机分为正常对照组、照射对照组和3个不同剂量EGP(100、200和400mg/(kg·d)]+照射组,按每10g体质量0.1mL灌胃,连续10d;正常对照和照射对照组均用等量生理盐水灌胃;7d后除正常对照组外,均进行一次性了射线2Gy照射,20h后检测巨噬细胞产生白细胞介素-1(IL-1)活性,以及血清白细胞介素-2(IL-2)、肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)水平和脾IFN-γ mRNA和蛋白表达量。结果(1)EGP200、400mg/kg+照射组小鼠巨噬细胞产生IL-1蛋白量(液闪计数,min^-1)分别为2347±254、3111±628,明显高于照射对照组的1327±729(P〈0.01);(2)EGP200、400mg/kg照射对照组小鼠血清102、TNF—α、IFN-γ(pg/mL)分别为31.62±1.71、124.27±17.28、110.57±15.71和34.30±2.14、136.19±17.94、123.57±14.47,明显高于照射对照组的23.06±1.79、99.76±9.41、88.28±10.62pg/mL(P〈0.05,P〈0.01);(3)EGP200、400mg/kg+照射组小鼠脾细胞IFN-γ耐州A和蛋白表达量(灰度值)分别为0.75±0.13、1.08±0.15、1.21±0.13和93247±5627、130154±7279、171467±8678,明显高于照射对照组的0.35±0.10、50096±4815(P〈0.01)。结论琼枝麒麟菜多糖(EGP)可以通过分泌上述细胞因子拮抗辐射损伤。

Abstract:Objective To investigate the effect of Euchemna gelatinae polysaccharide (EGP) on cytokine secretion in irradiated mice, so as to provide scientific basis for treatment of irradiated injury. Methods Fifty mice were randomly divided into 5 groups, normal control group, irradiated control group, and 3 EGP treatment groups which were treated with EGP 100,200,400 mg/(kg) respectively by intragastric injection for 10 days,and two control groups with equal volume of physiological saline. On the 7th d, the animals were irradiated with γ-rays (2.0 Gy). Some cytokines were examined 20 h after irradiation. The expression of mRNA and protein of interferon-γ (IFN-γ) in spleen cell were detected by RT-PCR and Western blot. Results (1) The level of interleukin-1 in macroghages in 200 mg EGP, 400 mg EGP groups (2 347 ± 2 540, 3 111 ± 628 min^- 1 respectively) were significantly higher than that in irradiated control group (1 327 ± 729, P 〈 0.01 ) ; (2)interleukin-2, tumor necrosis factor-α, and interferon-γin 200,400 mg EGP groups (31.62 ± 1.71,124.27 ± 17.28,110.57 ± 15.71 pg/mL; 34.30 ± 2.14,136.19 ±17.94,123.57 ±14.47 pg/mL respectively) were significantly higher than those in irradiated control group(23.06 ± 1.79,99.76 ± 9.41,88.28 ± 10.62 pg/mL, P 〈 0.05, P 〈 0.01 ) 3 ; (3) The expression of mRNA, and protein of IFN-γ in 3 EGP groups indicated by gray density (0.75 ± 0.13, 1.08 ± 0.15, 1.21 ± 0.13; 93 247 ± 5 627, 130 154 ± 7 279, 171 467 ± 8 678 respectively) were significantly greater than those in irradiated control group (0.35 ± 0.10; 50 096 ± 4 815, P 〈 0.01 ). Conclusion EGP may protect the mice against irradiation damage by enhancing the related cytokines secretion.
