薄荷油致小鼠肝毒性时-量关系及其机理研究

摘    要:目的研究薄荷油致小鼠肝毒性的量效、时效关系及自由基损伤机制。方法小鼠口服不同剂量薄荷油,测定其LD50。另取小鼠按不同剂量或不同时间点分组,检查血清ALT等肝功能指标和肝组织形态变化,测定肝组织SOD,MDA。结果小鼠口服薄荷油的LD50为3.0 ml/kg。ALT等肝功能指标在给药后24~48 h达到高峰,72 h可恢复近正常值。与正常组相比,薄荷油中、高剂量组对肝组织损伤显著,随着剂量增大,ALT等肝功能指标升高显著(P<0.05或P<0.01)。薄荷油组与正常组比较,小鼠肝组织中SOD降低而MDA升高(P<0.05),与四氯化碳相似。结论小鼠一次性口服大剂量薄荷油可造成急性肝损伤甚至死亡,并显示有毒性时-效、量-效关系;自由基损伤可能是其肝毒性机制之一。

Abstract:Objective To study the time-dose effects and free radical mechanism of peppermint oil on liver damnification in mice.Methods The medial lethal dose(LD50) of mice was determined when they took a different dose of peppermint oil orally.After other mice were divided into different groups according to different dose or time,hepatic functional parameters in serum were examined for example alanine aminotransferase(ALT) and so on.The morphologic change of hepatic tissues were observed,and superoxide dismutase(SOD) and malondialdehyde(MDA) of hepatic tissues were detected.Results The orally LD50 of peppermint oil was 3.0 ml/kg.Hepatic functional parameters in serum reached to the max at 24~48 hours after being intervened,which recovered close to normal values at 72 hours.Compared with that of the control group,hepatic tissues of the high and medium dose groups were damaged significantly,and hepatic functional parameters in serum advanced significantly(P<0.05 or P<0.01).SOD of hepatic tissues of mice taking peppermint oil was low and MDA of that was high compared with that of the control group,which was similar to that of carbon tetrachloride.Conclusion Acute toxicity even death may occur when mice take a large dose of peppermint oil once orally.Time-dose-toxicity effects would be indicated.Free radical damnification might be one of the liver injury mechanism of peppermint oil.
