湖南产不同种质、采收期、加工方法的山银花质量对比研究

摘    要:目的 比较湖南产不同种质、采收期和加工方法的山银花中主要有效成分含量的差异.方法 采用WatersSymmetry C18(150 mm×4.6 mm,5 μm)为色谱柱,流动相为0.4%冰醋酸-乙腈(梯度洗脱),流速为1.0 mL/min;高效液相色谱(HPLC)-紫外检测(UV) 法测定山银花中绿原酸的含量, 检测波长为330 nm;HPLC-蒸发光散射检测器(ELSD) 法测定此药材中灰毡毛忍冬皂苷乙和川续断皂苷乙的含量.结果 不同山银花样品中绿原酸含量为3.73%~8.23%,灰毡毛忍冬皂苷乙和川续断皂苷乙的总含量为4.16%~8.53%.其中,绿原酸含量和灰毡毛忍冬皂苷乙及川续断皂苷乙之和含量最高的是采自三青期的野生山银花,为8.23%、8.53%,微波干燥加工后的山银花中上述成分含量分别为7.05%、7.08%,其他加工方法含量均有不同程度降低.结论 野生山银花的质量优于栽培山银花,三青期采摘及微波干燥得到的山银花的质量最优,本试验结果可为山银花人工培育和采收加工提供实验依据.

Quality comparative study on different germplasm,harvest times and processing methods of Flos Lonicerae produced in Hunan

Abstract:Objective To compare the contents of the main effective components of different germplasm, harvest times and processing methods on Flos Lonicerae produced in Hunan. Methods The Waters Symmetry-C18 (150 mmx4.6 ram,5 μ m) was used with the mobile phase of 0.4% glacial acetic acid-acetonitrile (gradient elution), the flow rate was 1.0 mL/min, the detection wavelength was 33Onto. The content of chlorogenic acid in Flos Lonicerae was determined by HPLC-UV. The content of macranthoidin B and dipsacoside B was determined by HPLC-ELSD. Results The content of chlorogeneic acid in Flos Lonicerae was 3.73%-8.23%. The total content of macranthoidin B and dipsacoside B was 4.16%-8.53%. Among all samples, the content of ehlorogeneie acid, the total content of macranthoidin B and dipsacoside B in wild Flos Lonicerae picked in three Qing stage were highest, 8.23% and 8.53% respectively. Conclusion The quality of the wild Flos Lonicerae is better than the cultivated one, and Flos Lonicerae which is picked in three Qing stage and is dried by microwave has the best quality, which can provide the basis for artificial cultivation, harvesting and processing of Flos Lonicerae.
