| 摘 要： | 用激光扫描共焦显微镜观察了培养的小鼠腹腔巨噬细胞在ＣｏｎＡ刺激下的变化，结果显示ＣｏｎＡ刺激前溶酶体主要分布在细胞核周围，刺激后溶酶体在细胞周边明显增多，细胞外液中荧光强度增加．上述结果表明巨噬细胞在ＣＯｎＡ的刺激下，除形成吞噬小泡外，在刺激物加入后一定时间后，有外排小泡生成。在ＣＯｎＡ加入后的５ｍｉｎ内，细胞内的溶酶体的ｐＨ从静息状态的ｐＨ４．６上升到ｐＨ５．７，５ｍｉｎ后基本维持不变。

| Abstract: | Changes of cultured murine peritoneal macrophages stimulated by Con A are measured withLaser Scanning Confocal Microscope(LSCM).Aseries of images are obtained including phase- contrast im-ages,optical sectioning images,and 3- D reconstruction fluorescence images. The kinetic changes of lysoso-mal fluorescence intensity and pH are observed. These results show that the resting macrophage lysosomesare distributed in the perinuclear region. After stimulation by Con A for 30min, the number of lysosomesevidently increase at the cell periphery and the fluorescence intensity in the buffer also increases, It is sug-gested that Con A can induce macrophage endocytosis and exocytosis. The lysosomal pH rises from 4. 6 to5. 7 within 5 min after Con A is added,then basically keeps in the level of pH5. 7 for A period of time. |
