摘    要:目的:观察亚砷酸钠(Ars),内毒素(LPS)、肿瘤坏死因子(TNF)、烧伤血清、创伤血清对内皮细胞凋亡的影响及其机制。方法:体外培养内皮细胞ECV-304,与上述物质孵育24小时后检测凋亡水平及ICAM-1.bcl-2,p53表达的变化,结果:Ars可使内皮细胞发生剂量依赖的凋亡增加(与对照相比,P<O.01),但是LPS、TNF烧伤血清和创伤血清对内皮细胞的作用有限,细胞凋亡率在15%以下。除10%烧伤血清外,所有处理因素都内皮细胞ICAM-1表达降低。体外诱导内皮细胞凋亡与bcl-2表达降低、p53表达增多有关,结论:Ars,LPS,TNF烧伤血清和创伤血清通过下调bcl-2表达,上调p53表达来影响内皮细胞凋亡,并且抑制内皮细胞功能。

Abstract:Objective: To investigate the endothelial cell apoptosis inducing effect of substances, including sodium arsenite (Ars), lipopolysaceharide (LPS), tumor necrosis factor (TNF), burned serum and traumatic serum and the mechanisms involved. Methods: By comparing the changes of apoptosis rates and the expressions of ICAM -1, bc1 - 2, p53 after endothelial cells ECV - 304 were incubated with these apoptosis inducing substances. Results: Ars increased the apoptosis rate of endothelial cells and this effect was dose-dependent (compared with control, P<0. 01. LPS, TNF, burned serum and traumatic serum had only limited effect on endothelial cell apoptosis, the apoptosis rate was below 15% Except for 10% burned serum, all these substances decreased the expression of endothelial cell ICAM - 1, in vital en- dothelial cell apoptosis inducing effect were correlated to the decrease in the expression of bcl - 2 and the increase in the expression of p53 and ICAM - 1. Conclusion: Ars, LPS, TNF, burned serum and traumatic serum influenced endothelial cell apoptosis through down - regulation of bcl - 2 and up - regulation of p53 and inhibited their functions.
