低频重复经颅磁刺激对帕金森病大鼠行为学的影响

摘    要:目的探讨0.5Hz重复经颅磁刺激(rTMS)对帕金森病大鼠行为学的影响。方法取健康SD大鼠45只,用6-OHDA毁损右侧黑质致密部(SNc)与中脑腹侧背盖(VTA)制备偏侧帕金森病模型。用阿扑吗啡(APO)腹腔注射筛选,将成功的20只帕金森病鼠随机分为治疗组和对照组,每组10只。治疗组给予rTMS治疗,频率为0.5Hz,刺激强度为阈上30%,连续20个刺激,每天1次,连续刺激2周。对照组予以假刺激。在治疗前、治疗结束后第1天、14天、28天分别测定APO诱导的大鼠旋转圈数、启动潜伏期的变化。结果(1)大鼠旋转圈数(r/min):治疗组与对照组在治疗结束后第14天为(9.7±3.0vs14.6±2.6)、28天为(9.2±2.7vs14.2±2.6),治疗组旋转圈数减少,与对照组比较差异有统计学意义(P<0.05);治疗组在治疗结束后第14天、28天与治疗前比较差异有统计学意义(P<0.05);对照组各时间点比较差异无统计学意义(P>0.05)。(2)启动潜伏期(s):治疗组与对照组rTMS治疗结束后第1天(25±9vs39±16)、第14天(18±7vs42±16)、第28天(17±7vs42±16)比较,治疗组启动潜伏期明显缩短,差异有统计学意义(P<0.05);治疗组各时间点与治疗前比较差异有统计学意义(P<0.05);对照组各时间点比较差异无统计学意义(P>0.05)。结论0.5Hz、阈上30%刺激强度的rTMS可改善PD大鼠的行为学异常。

Effects of low frequency and repetitive transcranial magnetic stimulation on behavior of rats with Parkinson's disease

Abstract:Objective To study the effects of repetitive transcranial magnetic stimulation (rTMS) on behavior of rat with Parkinson's disease. Methods The Parkinson's disease model was built in 45 healthy SD rats, which were destroyed the right substantia nigra pars compact(SNe) and ventral tegrnental area(VTA) with 6-OHDA. And then injected apomorphine (APO) intraperitoneally, to provide 20 successfully PD models rats. All PD rats were divided into control and treatment groups randomly, 10 rats in each group. The treatment group was given the stimulation, which intensity was 30% over threshold, the frequency was 0.5Hz, 20 stimulates successive, once a day, 2 weeks duration. While the control groups were given false stimulation. The numbers of rotation induced by APO and initiative latent period were observed before treatment. the 1 st day, 14th day and 28th day after treatment. Results ( 1 ) The rotation numbers (r/min) of treatment group in the 14th day (9.7 ± 3.0 vs 14.6 ± 2.6) and 28th day(9.2 ± 2.7 vs 14.2 ± 2.6) after treatment were significantly decreased comparing with that of control group ( P 〈 0.05). And the results of treatment group were significant different before and 14th day, 28th day after treatment ( P 〈 0.05) ,while no statistical significance in control group( P 〉 0.05). (2)The initiative latent period (s) in treatment group significantly shortened at days 1st (25 ± 9 vs 39 ± 16) ,14th ( 18 ± 7 vs 42 ± 16) and 28th( 17 ± 6 vs 42 ± 16) after rTMS than that in the same time point in control group ( P 〈 0.05). And the period m experimental group significantly shortened at days 14th and 28th after rTMS than that before treatment( P 〈 0.05). There was no significantly difference before and after treatment in control group( P 〉 0.05). Conclusion It proved that the rTMS with 0.5Hz, 30% over threshold could improve the behaviors disorder in PD rats.
