肝细胞生长因子在慢性肾衰竭大鼠肾组织中表达的研究

摘    要:目的研究肝细胞生长因子(HGF)对慢性肾衰竭大鼠肾组织中基质金属蛋白酶2(MMP-2)及其抑制剂(TIMP-2)表达的影响,探讨HGF延缓肾衰竭进程的可能机制。方法给大鼠腺嘌呤250mg/(kg·d)灌胃建立慢性肾衰竭模型;对照组用同体积2%淀粉溶液同时间灌胃。免疫组化检测正常对照组、慢性肾衰竭模型组HGF.MMP-2及TIMP-2的表达。结果较正常对照组,各时间点模型组肾脏病理损害进行性加重;免疫组化示,灌胃第7天,HGF、MMP-2表达明显增加至第14,21天,HGF、MMP-2表达缓慢下降;TIMP-2持续上升;HGF表达与MMP-2表达呈正相关,与TIMP-2呈负相关。结论HGF抗肾间质纤维化作用可能与MMP-2、TIMP-2密切相关。

Abstract:Objective To investigate the effect of hepatocyte growth factor(HGF) on the expression of matrix metalloproteinase- 2(MMP-2) and its tissue inhibitors of metalloproteinase-2 (TIMP-2) in renal tissue of rats with experimental chronic renal failure, and study the possible mechanism of antifibrotie effects of HGF. Methods The rats in adenine-induced model group received :250 mg/( kg.d) by gavage with 2.5% adenine and 2% starch solution. The rats in control group only received starch solution by gavage at the same time. Haematoxylin and eosin(HE) and Masson staining were used to study the dynamic histological changes of the kidneys. The expressions of HGF, MMP-2,TIMP-2 were analyzed by immunohistochemistry staining. Results Renal damages were exacerbated in model group in comparison with those of normal control group at each time point. The expression of HGF was positive related to the expression of MMP-2 and negatively correlated with the expression of TIMP-2. Conclusion The mechanisms of antifibrotie effects of HGF may be associated with MMP-2 and TIMP-2 in renal interstitial progress.
