肉苁蓉总苷体外清除自由基及对OH·引发的DNA损伤的保护作用

摘    要: 目的研究肉苁蓉总苷体外清除自由基作用及对OH·引起的DNA损伤的保护作用。方法采用现代生物化学发光技术,检测肉苁蓉总苷对O2,OH·,H2O2等自由基的清除作用。并通过Cu2-Phen-H2O2-VitC-DNA发光体系研究肉苁蓉总苷对OH.引起的DNA氧化损伤的保护作用。结果肉苁蓉总苷对O2,OH·,H2O2活性氧自由基均有清除作用。其50%抑制浓度( IC50)分别为0.0731,7.031,0.098,0.1254mg·mL-1。对DNA化学发光的IC50为0.4211ug·mL-1。结论实验结果表明,肉苁蓉总苷能有效地清除各种活性氧自由基,能保护OH·引发的DNA氧化损伤。

Abstract:OBJECTIVE The extrasomatic free radical scavenging effects of glycosides of Crstanche (GCs) and its protective effects against OH·-induced DNA damage were explored. METHODS The free radical scavenging effects of GCs were measured by Chemiluminescence methods and the protective effects against OH·-induced DNA damage were measured with Cu2+-phen-H2O2-VitC-DNA Chemiluminescence system.RESULTS GCs inhibited the O2,OH·,H2O2 and their 50% of inhibitory concentration(IC50) were 0.0731,7.031,0.098,0.1254mg.·mL-1, respectively. IC50 for DNA Chemiluminescence intensity of GCs was0.4211ug·mL-1. CONCLUSIONS GCs could scavenge free radicals effectively and has protection against OH·-induced DNA damage.
