摘    要:目的 探讨包裹抗癌药物表柔比星(EPI)的PEG化壳聚糖纳米粒(PEG/CS-EPI NPs)对5-8F鼻咽癌细胞株的增殖抑制作用。方法 应用阴离子凝聚法制备PEG/CS NPs,并负载EPI,激光粒度分析仪测定其粒径,透射电镜观察其形态特征,通过MTT法测定和研究PEG/CS-EPI NPs对5-8F鼻咽癌细胞株的体外增殖抑制作用。结果 PEG/CS-EPI NPs呈圆形或椭圆形,粒径分布均匀,平均粒径322.1nm,对5-8F细胞株的增殖抑制作用具有浓度和时间依赖性,随药物作用时间的延长和浓度的增大,抑制率逐渐提高。结论 PEG/CS-EPI NPs性能良好,制备工艺简单,有望成为化疗药物的新型制剂。

Inhibition of the proliferation of nasopharygeal cells by PEG-chitosan nanoparticles loaded with epirubicin
Abstract:Objective To investigate the effect of PEG-chitosan nanoparticles loaded with epirubicin (PEG/CS-EPI NPs) on cell proliferation in nasopharygeal cell line 5-8F. Methods The ionic gelation technique was employed to prepare the PEG/CS-EPI NPs and their characters were detected by TEM and photo correlation spectroscope(PCS); the loaded capacity was measured by ultraviolet spectrophotometry; The cytotoxicity of PEG/CS-EPI NPs and proliferation of 5-8F cells were detected by MTT assay. Results The EPI-loaded PEG/CS NPs were discrete and uniform spheres with average diameter 322.1 nm. It had a sustained antitumor activity against 5-8F cells in a time- and dose-dependent manner. Conclution The PEG/CS-EPI NPs with a good property of drug sustained release and an easy process of preparation, promises to be a novel sustained release system for chemotherapy drugs.
