| 摘 要： | 的 探讨1,25-二羟维生素D3[1,25(OH)2D3]与顺铂联合对人卵巢癌细胞株HO8910的增殖抑制情况,并测定二者作用下S期激酶相关蛋白(Skp2)及肿瘤抑制基因p27的蛋白表达情况,进一步探讨它们的作用机制.方法 用四唑盐(MTT)比色法测定1,25(OH)2D3、顺铂及二者联合对HO8910细胞株作用后细胞增殖情况,流式细胞仪进行细胞周期分析,免疫细胞化学染色检测Skp2及P27蛋白表达情况.结果 与对照组(未用药)相比,1,25(OH)2D3和顺铂作用下细胞吸光度(OD)减少,细胞增殖受到抑制(P<0.01),二者联用时此作用增强,二者作用下G0/G1期细胞增多,Skp2表达减少、P27蛋白表达增多(P<0.01).结论 1,25(OH)2D3有加强顺铂抑制肿瘤细胞增殖的作用,二者均可能通过下调Skpz表达、上调P27蛋白表达,从而抑制癌细胞增殖.
