脊髓损伤后水通道蛋白-4的时程变化

摘    要:探讨脊髓损伤后水通道蛋白-4及其mRNA的变化规律,应用改良Allen脊髓损伤的模型,于术后1、3、7和14 d取脊髓组织,采用免疫组织化学和原位杂交技术检测AQP-4的表达。结果表明,AQP-4及其mRNA表达丰富的部位主要集中在损伤脊髓的后索、前索和后角,以小胶质细胞和少突胶质细胞为主。AQP-4及其mRNA表达在脊髓损伤后1 d开始增加,在脊髓损伤后3 d达到高峰。随后下降,对应的病理改变为细胞内水肿。本研究结果提示:胶质细胞AQP-4表达增强与早期损伤性脊髓水肿密切相关,它可能是形成早期细胞内水肿的重要因素之一。

Abstract:To study the time course changes of aquaporin(AQP)-4 and AQP-4 mRNA after spinal cord injury, Allen's model of spinal cord injury was set up. Spinal cords were removed and fixed at 1 d,3 d,7 d and 14 d after spinal cord injury, and examined by immunohistochemistry and in situ hybridization. Expression of AQP-4 protein and mRNA of oligodendrocyte and mi-crogliocyte in spinal cord injury was predominantly observed in posterior funiculus, anterior funiculus and posterior horn. It started to increase at 1 d after spinal cord injury and reached the peak at 3 d, -and then decreased at 7 d. The corresponding histo-logical changes at this stage were intracellular edema. These results suggest that up-regulation of AQP-4 in glial cells has a close relationship with the early stage of the traumatic spinal cord edema and may play an important role in the intracellular edema.
