摘    要:目的分析对比三种染色方法在显示新生骨成骨情况的差异性,为体内组织工程研究提供更好的组织学鉴别方法。方法将大节段多孔生物陶瓷/聚乳酸复合支架植入犬4个不同的非骨部位,6个月后取材脱钙,石蜡包埋及切片分别做苏木精-伊红染色、Masson染色和亚甲基蓝-碱性品红染色,在光镜下观察其组织学形态并做比较。结果苏木精-伊红染色下可以清楚的观察到成骨细胞、类骨质等成分;Masson染色胶原纤维及成骨现象着色鲜艳,易于观察;亚甲基蓝-碱性品红染色成骨细胞更加清晰,与周围组织对比明显。结论苏木精-伊红染色更符合常规病理学观察习惯,Masson染色和亚甲基蓝-碱性品红染色在胶原纤维、成骨细胞等成分的观察上更具优势,因此三者结合更有利于镜下组织形态学观察。

The histological study on bone tissue engineering by defferent stainings

Abstract:Objective To compare three different staining methods for osteogenesis,and to provide better histological method for the study of in vivo tissue engineering.Methods Implanted large segment of porous bio-ceramic in four different non-bone parts of dogs.Drawn biopsies from there after 6 months.Made paraffin sections after decalcified them.Stained the sections by HE,Masson and methylene blue-basic fuchsin stainings.Then observed their histological patterns by microscope,and compared the stainings.Results The osteoblasts and osteoid could be observed easy in the sections stained by HE staining.The collagen fiber and osteogenesis was well stained and could be easy to observe in the sections stained by Masson staining.Compared to the surrounding tissue,the osteoblasts were easier to observe in the sections stained by methylene blue-basic fuchsin staining.Conclusion HE staining was generally used in pathology.The osteoblasts and collagen fiber were more obviously in Masson and methylene blue-basic fuchsin staining.So the combination of this 3 stainings would be more favorable for the microscopically observation of morphological patterns.
