龟板对脊髓损伤后脊髓雌激素α受体及其基因表达的影响

摘    要:目的 :观察龟板对脊髓损伤后脊髓雌激素α受体 (ER -α)及其基因表达的影响。方法 :以改良的Allen法建立大鼠脊髓损伤模型 ,随机分为对照组、模型组和龟板治疗组 ,用免疫组织化学和原位杂交方法检测脊髓中ER -α及其mRNA的表达。结果 :模型组中脊髓ER -α受体及其mRNA的阳性细胞数与对照组比较显著减少 ;治疗组中ER -α及其mRNA的阳性细胞数与模型组比较显著增加。结论 :龟板可增加脊髓损伤后脊髓ER -α及其基因的表达。

Abstract:OBJECTIVE:To study the effect of Plastrum Testudinis on the ER-αand its mRNA in the spinal cord folˉlowing spinal cord injury.METHODS:The model was established with modified Allen spinal cord injury method.Rats were divided into three groups:the control group,spinal cord injury group,Plastrum Testudinis group.The changes of the ER-αand ER-αmRNA positive cells in the spinal cord was studied by immunohistochemistry and in situ hybridization.RESULTS:The number of ER-αand ER-αmRNA positive cells in the spinal cord motor neuron of the spinal cord injury group was less than that of the control group;Plastrum Testudinis increased the number of ER-αand ER-αmRNA positive cells in the spinal cord motor neuron of the spinal cord injury rats.CONCLUSION.Plastrum Testudinis could increase the expression of ER-αand ER-αmRNA in the spinal cord motor neuron to enhance estrogen neuroprotection activity.
