动脉粥样硬化性脑梗塞患者血脂与对氧磷酶基因多态性的关系

摘    要:目的:研究动脉粥样硬化性脑梗塞患者血脂水平与对氧磷酶(PON_1)基因192位点多态性的关系。方法:应用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)方法,对动脉粥样硬化性脑梗塞52例,正常人48例的PON_1基因192位点多态性进行分析,并同时检测血脂水平。结果:与正常组相比脑梗塞组存在明显的血脂异常(P<0.05),两组人群PON_1基因192位点基因型分布处于Hardy-Weinberg平衡,且脑梗塞组R等位基因频率明显高于正常组,分别为0.65、0.48(P<0.05)。但血脂水平与各基因型之间比较无显著差异(P>0.05)。结论:PON_1基因192位点多态性可能与动脉粥样硬化性脑梗塞有关,但未发现其与血脂水平有关。

Abstract:Objective:To explore the relationship between the paraoxonase(PON1)gene 192 polymorphism and the level of serum lipids in patitents with atherosclerotic cerebral infarction(ACI). Method:Using polymerase chain reaction (PCR)-restrict fragment length polymorphism(RLFP) ,We assayed the genotype and allele frequency of PON1 gene 192 polymorphism in 52 patients with ACI and 48 healthy control subjects. Sera lipids were measured at same time. Results: The level of sea lipids in ACI group were significantly abnormal compared with those in control group. The distribution of genotypes in the two groups was at the Hardy-Weiberg equilibriums. But the frequency of the R allele of PON1 gene in ACI group was significantly higher than that in control group. (0. 65 vs 0. 48. P<0. 05) But no significant differences were found among the level of sera lipids in ACI group for PON] gene 192 in different genotypes. Conclusion: The PON1gene 192 polymorphism should be associated with ACI to some extent in Chinese. B allele of PON1 gene could be a relative risk factor in ACI . But the paraoxonase (PON1) gene 192 polymorphism was not associated with the level of sera lipids in patitents with atherosclerotic cerebral infarction (ACI).
