K-ras基因与c-myc基因在大肠癌中的表达及相关性研究

摘    要:目的 通过研究k-ras基因与c-mye基因在大肠癌中的表达情况,初步探讨二者在大肠癌发生、发展过程中的作用及可能的分子机制.方法 采用S-P免疫组织化学方法 检测80例结直肠癌组织及15例正常大肠黏膜中k-ras基因与c-myc基因的表达情况.结果 大肠癌组织中k-ras阳性表达率65.0%(52/80),c-myc阳性表达率52.50%(42/80).k-ras基因与c-myc基因在正常组织中未见表达,在大肠癌组织中呈阳性表达.随着分化程度的降低k-ras基因与c-myc基因的阳性表达率也明显增高(P<0.05);k-ras突变与年龄、性别、有无淋巴结转移无相关性.c-mye阳性表达与大肠癌的临床分期、淋巴结转移密切相关(P<0.05);k-ras基因与c-myc基因之间存在明显的相关性(P<0.05).结论 k-ras基因在大肠癌中的表达与细胞分化程度、Dukes分期有密切关系;c-myc在大肠癌中的表达与细胞分化程度、Dukes分期及有无淋巴结转移有密切关系;而k-ras基因与c-myc基凼的表达与年龄、性别无明显关系.k-ras基凼与c-myc基因的表达存在明显的止相关关系.

Expressions and correlation of k-ras and c-myc in colorectal carcinoma
Abstract:Objective To examine the expressions of k-ras and c-myc in colorectal carcinoma.and to analyze their correlation.Methods The expressions of k-ras and c-myc proteins were detected in 80colorectal carcinoma samples and 15 normal mucosas by immunohistochemistry S-P method,and their correlation was then analyzed.Results The positive expressions of k-ras was 65.0%(52/80),and Cmyc was 52.50%(42/80).k-ras and c-myc were rarely seen in normal organism.but were remarkably decreased in colorectal carcinoma tissues.k-ras and c-myc expressions were inverse correlation to cell differentiation(P<0.05);k-ras catastrophe was no related with age,sex and lymph node metastasis.The positive expression rate of c-myc was related with Dukes stage and lymphatic metastasis of colorectal carcinoma(P<0.05).There is obviously correlation between k-ras and c-myc(P<0.05).Conclusion The expressions of k-ras was related with cell differentiation,Dukes stage;The expressions of c-myc is significantly related with cell differentiation,Dukes stage and lymphatic metastasis of colorectal carcinoma.But there is no correlation with age and sex.The expressions of k-ras and c-myc direct correlation each other in colorectal carcinoma.
