体外诱导脐带血干细胞成心肌细胞的实验研究

目的 :为探讨体外诱导脐带血干 /祖细胞形成心肌细胞的可行性以及寻找最佳诱导条件。方法 :取胎盘的脐带血经Ficoll处理后提取单个核细胞层 (MNC) ,在不同浓度的 5 -aza的DMEM培养基中诱导培养 ,用心肌特有的肌球蛋白重链 (MHC)作免疫组化标记诱导形成的心肌细胞。结果 :用 5 -aza体外诱导培养能形成心肌细胞 ,仅1 0 μmol/L形成的心肌细胞最多。 结论 :脐带血在体外培养条件下经 5 -aza处理能诱导分化成心肌细胞 ,最佳浓度为 1 0 μmol/L。脐带血可作为修复损伤心肌的细胞移植的一个来源

The Study of Forming Cardiomyocytes by Ducing Stem Cells of Cord Blood in vitro

Objective To investigate the feasibility of forming cardiomyocytes by inducing stem cells/progenitor cells of cord blood (CB) in vitro and to seek out the best inducing conditions. Methods Taking mononuclear cell (MNC) from CB of the placenta by Ficoll to induce and culture in the different concentrated culture medium of 5-aza DMEM. The cultured cells were immunohistochemically stained for myosin heavy chain (MHC). Result The cultured cells with 5-aza formed myotubules which stained positively for myosin heavy chain.Conclusions The CB can be induced and differentiated into cardiomyocytes by 5-aza in vitro . The best concentration was 10μmol/L?Umbilical cord blood may be a source of cell transplantation?
