当归对单侧输尿管梗阻大鼠肾组织中黏附分子ICAM-1表达的影响

摘    要:目的:观察当归对单侧输尿管梗阻(UUO)大鼠肾小管间质纤维化过程中黏附分子P-选择素(P-selectin)和细胞间黏附分子-1(ICAM-1)、转化生长因子-β1(TGF-β1)等细胞因子的表达和CD68单核/巨噬细胞浸润的影响。方法:采用大鼠模型,52只SD大鼠分为:假手术组、UUO组、当归组。术后第7d和第14d留取标本处死大鼠,用免疫组化方法检测肾组织内P-selectin、ICAM-1、CD68、TGF-β1的表达。结果:与假手术组相比,UUO组大鼠第7d和第14d肾组织内ICAM-1、P-selectin、CD68、TGF-β1的表达均显著上调(P<0.05)。与UUO组相比,当归组两个时间段肾小管间质病变减轻,且肾组织内ICAM-1、CD68、TGF-β1的表达受到显著抑制(P<0.05)。结论:当归可能是通过抑制ICAM-1的表达和炎细胞的组织浸润,从而降低了肾小管间质TGF-β1的水平和纤维化的进展。

Abstract:Objective: To investigatethe effect of angelica on the expression of P-selectin,Intercellular Cell Adhesion Molecule-1(ICAM-1),and Transforming Growth Factor-1(TGF-β1),and the infiltration of macrophage in kidney of UUO rats.Methods: the unilateral ureteral obstruction(UUO) rat model was studied in this experiment.52 male SD rats were randomly divided into: ① sham group;② UUO model group;③Angelica group.Half rats in each group were killed respectively on day 7 and day 14.Renal tissue P-selectin,ICAM-1,CD68,TGF-β1 expression and localization were detected by immmunohistochemical analysis.Results: Compared with the sham group,the levels of renal tissue ICAM-1,P-selectin,CD68,TGF-β1 expression were significantly upregulated in UUO model group on day 7 and day 14(P<0.05).Angelica decreased tubulointerstitial injury score from(5.56±0.56) to(4.42±0.89),and the renal interstitial inflammatory cell count,and suppressed significantly the levels of renal tissue ICAM-1,CD68,TGF-β1 expression on day 7 and day 14,compared with the UUO model group(P<0.05).Conclusion: Angelica meliorates development of tubulointerstitial fibrosis via decreased ICAM-1 expression and inflammatory cell infiltration in UUO rats.
