摘    要:目的探讨早期生长反应因子1(early growthresponse-1,Egr-1)在经H2O2损伤后的晶状体上皮细胞中的表达及其意义。方法用SP免疫组化法测定经H2O2处理后2h、8h、24h的晶状体上皮细胞中Egr-1的表达,用MAIS300图像分析系统测定其光密度值,采用t检验分析其在各时间点的表达水平。结果在H2O2作用后2h的晶状体上皮细胞中,Egr-1多为强阳性表达,平均光密度值为0.55±0.08;在H2O2作用后8h的晶状体上皮细胞中,Egr-1多为阳性表达,平均光密度值为0.40±0.09;两组比较,差异有非常显著的统计学意义(P〈0.01)。在H2O2作用后24h的晶状体上皮细胞和对照组晶状体上皮细胞中,Egr-1未见表达,平均光密度值为0。结论Egr-1在H2O2损伤后的晶状体上皮细胞中早期表达,可能为氧化损伤引起的白内障的早期反应之一。

Abstract:Objective To explore the expression and significance of early growth response-1 (Egr-1) in lens epithelial cells impaired by hydrogen peroxide (H2O2). Methods The Egr -1 expression of lens epithelial cells impaired by H2O2 was detected by immunohistochemistry (SP assay). Results Two hours after treatment with H2O2, the expression of Egr-1 in the epithelial cells was hard positive; opacity density value was 0.55±0.08; 8 h after treatment with H2O2, the expression of Egr-1 in the epithelial cells was positive and the opacity density value was 0.40 ±0.09; the difference was significant. While 24 h after treatment with H2O2, the expression of Egr -1 was negative in lens epithelial cells and in the control group. Conclusion Egr -1 is expressed in epithelial cells in the early stages after treatment with H2O2, and this may be one of the early activity in the development of hydrogen peroxide -induced cataract.
