波长510.6 nm铜蒸气激光诱导兔血管平滑肌细胞凋亡的照射剂量筛选

摘    要:目的 筛选对离体培养的兔血管平滑肌细胞 (VSMC)能诱导出较高凋亡率的激光照射剂量参数。方法 取离体培养的第 4代兔VSMC ,予波长 5 10 6nm铜蒸气激光照射 ,照射剂量为功率密度 10 0、2 0 0、30 0mW cm2 ,能量密度10 0、15 0、180、2 0 0、2 10、30 0J cm2 。脱氧核苷酸末端转移酶介导的缺口末端标记法 (TUNEL)标记检测各照射剂量组细胞凋亡率。结果 与对照组比较 ,各照射组VSMC凋亡率明显增高 ,达对照组凋亡率的 2 9~ 14 7倍 (P <0 0 5 )。以功率密度 30 0mW cm2照射 6 6 7s照射组之VSMC凋亡率最高 ,达 30 %。结论 以波长 5 10 6nm、功率密度 30 0mW cm2 铜蒸气激光照射兔VSMC 6 6 7s可诱导发生较高的细胞凋亡率

Abstract:Objective To select an optimal laser dosage of 510 6 nm laser irradiation which could induce apoptosis of vascular smooth muscle cells (VSMC). Methods The rabbit VSMC cultured in vitro were irradiated by 510 6 nm copper vapor laser with a power density of 100, 200 or 300 mW/cm 2 and an energy density of 100, 150, 200, 210, 300 J/cm 2, respectively. TUNEL staining was used to label apoptotic cells of VSMC irradiated at different dosages. Results Apoptotic rate in all laser irradiation groups was 2 9 to 14 7 times higher than that of control group(P<0 05). The apoptotic rate of VSMC irradiated at 300 mW/cm 2 for 667 s was 30%. Conclusion Apoptotic rate of rabbit VSMC irradiated at 300 mW/cm 2 for 667 s by 510 6 nm copper vapor laser was relatively higher.
