摘    要:目的构建针对乙肝病毒S基因的RNAi表达载体,观察其对HBV表达的影响。方法合成针对HBVS基因的RNAi寡核苷酸,克隆入pSUPER载体,与pHBV质粒共转染HepG2细胞。ELISA和Westernblot检测HBsAg表达。结果成功构建针对HBVS基因的RNAi表达载体pSUPER-S1和pSUPER-S2对HBsAg的抑制率分别为79.0%和43.2%,无关序列的RNAi表达载体无此作用。结论载体介导的RNAi能高效、特异地抑制HBVS基因的表达。

Inhibitory efects of vector-based RNAi on S gene expression of hepatitis B virus

Abstract:Objective To construct RNAi expressing vectors targeting HBV S gene and to evaluate the inhibitory effects on HBV replication.Methods Two sequences targeting HBV S gene were cloned into pSUPER vector and then were cotransfected with pHBV into HepG2 cells.HBsAg expression was assayed by ELISA and Western blot.Results PSUPER-S1 and pSUPER-S2 RNAi expressing vectors were successfully constructed and inhibited HBsAg in HepG2 cells by 79.0% and 43.2%,Whereas irrelevant RNAi expressing vector did not show inhibitory effects on the HBsAg expression.Conclusion Vector-based RNAi can inhibit HBV replication effectively and specifically.
