基质金属蛋白酶-9和原癌基因c-myc在甲状腺癌中的表达及其关系探讨

目的探讨基质金属蛋白酶-9（MMP-9）和原癌基因c-myc在甲状腺癌中的表达及其关系。方法手术中获得的甲状腺癌、甲状腺腺瘤、桥本氏甲状腺炎及正常甲状腺组织的新鲜标本共39例，提取其总RNA，然后进行半定量逆转录多聚酶链反应（RT—PCR），比较MMP-9及c—myc mRNA在不同疾病组织中的表达差异。用酶联免疫吸附方法（ELISA）检测不同患者血清中的MMP-9水平。结果在甲状腺癌组织中，MMP-9及c—myc mRNA的表达均显著性高于甲状腺腺瘤、桥本氏甲状腺炎及正常甲状腺组织（P〈0．05），其他3组问的比较无统计学意义。甲状腺癌患者同时伴有血清MMP-9水平的显著性升高。结论MMP-9及c—myc mRNA的表达增高及血清MMP-9水平的升高可能与甲状腺癌的发生有关。

Expression of matrix metalloproteinase 9 (MMP-9) and c-myc genes in thyroid carcinoma

[Objective] To investigate the expression of matrix metalloproteinase 9 (MMP-9) and c-myc genes in thyroid carcinoma and their relation in the pathogenesis of thyroid carcinoma. [Methods] The thyroid tissues were selected in surgical operation, including 9 thyroid carcinoma cases, 8 Hashimoto's thyroiditis cases, 11 thyroid adenoma cases and 11 normal tissues. The total RNA was extracted and gene expression of MMP-9 and c-myc was evaluated by RT-PCR. The serum MMP-9 levels were determined with ELISA assays. [Results] The levels of MMP-9 and c-myc mRNA in thyroid carcinoma group were significantly higher than those in other 3 groups (P <0.05). Also, the serum MMP-9 levels in patients with thyroid carcinoma were significantly increased. [Conclusions] The high expression of MMP-9 and c-myc mRNA and serum MMP-9 levels appear to be related with the pathogenesis of thyroid carcinoma.
