维拉帕米对肝星状细胞增殖和凋亡的影响

摘    要:目的观察维拉帕米对人肝星状细胞(hepatic stellate cells,HSCs)增殖和凋亡的影响。方法采用不同浓度的维拉帕米对HSCs处理48 h,CCK-8试剂盒(cell counting Kit-8)检测细胞增殖抑制率,HE染色观察细胞形态超微结构的变化,流式细胞仪Annexin-V/PI双染法检测HSCs的凋亡率。结果维拉帕米可抑制HSCs的增殖,且有剂量相关性,其IC50值为(150±10)mmol·L-1;HE染色可见维拉帕米组出现不同程度的细胞体积变小、核浓缩深染等凋亡形态改变;Annexin-V/PI双染法检测显示,不同浓度的维拉帕米均可诱导HSCs凋亡,与阴性对照组比较,差异有统计学意义(P<0.05)。结论维拉帕米可抑制HSCs的增殖,诱导其凋亡。

Abstract:Objective To observe the effects of verapamil on proliferation and apoptosis of human HSCs.Methods HSCs were incubated with different concentration of verapamil,cell proliferation was assessed by cell counting Kit-8,morphologic change was detected by HE staining,the rate of apoptosis was detected by flow cytometry with Annexin-V/PI staining.Results Verapamil could inhibit the proliferation of HSCs in a dose-dependent manner,the IC50 value was(150±10)mmol · L-1.HE staining showed different changes of cell apoptosis such as soma shrinking,nucleus concentrating and anachromasis,and so on.The apoptosis of HSCs was induced by different concentration of verapamil,which was different from the negative control group(P<0.05).Conclusions Verapamil can inhibit the proliferation of HSCs and induce their apoptosis.
