+GZ暴露对大鼠脑热休克蛋白-70表达水平的影响

摘    要:目的 探讨+Gz暴露条件下大鼠脑中热休克蛋白-70(HSP70)的变化及其在+Gz所致脑损伤中的作用。方法 100只大鼠,随机分为4胡,每组25只,一组作为空白对照,其余三组分别进行+2Gz,+6Gz和10Gz暴露,峰值作用时间3min,加速度增长率1G/s然后,分别于暴露后的6h、1d、2d、4d和6d麻醉大鼠,心脏灌注后迅速取脑组织,用West blot法比较HSP70的变化情况。结果 +Gz暴露后6hHSP70开始升高,1d达到高峰,至6d时基本恢复正常。不同+Gz暴露之间进行比较可以看出,+6GGz组HSP70表达最高,+10Gz最少。结论 +Gz暴露对大鼠脑HSP70表达水平有显著的影响。

Abstract:Objective To explore the effect of G Z exposures on expression of heat shock protein 70(HSP70) in the rat brain. Methods One hundred SD rats were randomly divided into 4 groups: 1 control group, and 3 experimental groups, which underwent 2 G Z, 6 G Z, 10 G Z exposures for 3 min respectively. The expression levels of HSP70 in rat brain were measured by West blot 6 h, 1 d, 2 d, 4 d and 6 d after G Z exposures. Results As compared with control, the expression levels of HSP70 began to raise 6 h after G Z exposure, up to the peak value 1 d later, and returned to normal after 6 d. As to the degree of change of HSP70 expression following various G Z exposures, the most obvious elevation was observed after 6 G exposure, and the least after 10 G exposure. Conclusion Increase of HSP70 expression in rat brain can be induced by G Z exposures. It may play a role in protecting the brain from injury following G Z exposures.
