摘    要:目的:探讨HDGF基因特异性siRNA对肝癌细胞系HepG2的增殖能力及ADAM9基因表达的影响.方法:设计并合成HDGF基因特异性的siRNA干扰序列,瞬时转染HepG2细胞.实时荧光定量PCR检测HDGF和ADAM9 mRNA的表达变化;MTT法观察细胞增殖能力的变化.结果:转染48h时HDGF和ADAM9的基因表达抑制最为明显,分别为阴性对照组表达量的7%和5%(P<0.05).转染48h时HepG2细胞增殖能力受到明显抑制(P<0.05).结论:HDGF siRNA能够显著抑制HDGF和ADAM9基因的表达,降低HepG2细胞的增殖能力.

Abstract:Objective: To study the effect of siRNA targeting hepatoma - derived growth factor (HDGF) on prolif- eration of the hepatoma cell line HepG2 and expression of metalloprotease 9 (ADAM9). Methods: Specific siRNA for HDGF gene was designed and synthesized, followed by transient transfection into the HepG2 cells. RT- PCR was used to examine the expression of HDGF and ADAM9. The proliferation of the HepG2 cells was examined by MTT assay. Results :The expression of HDGF and ADAM9 was found to be significantly suppressed at 48 hour (decreased to 7% and 5% respectively, P 〈 O. 05 ), and the proliferation of the HepG2 cells was also suppressed significantly at 48 hour (P 〈 0.05 ). The expression of ADAM9 was positively correlated with the expression of HDGF, significantly sup- pressed at 48 hour. Conclusion: HDGF siRNA can significantly silence the expression of HDGF and ADAM9,inhibit the proliferation ability of the HepG2 cells.
