稳定表达NT-3的NIH3T3细胞对小鼠耳蜗螺旋神经节细胞生长的影响

摘    要:目的通过体外共培养观察NT-3对耳蜗螺旋神经节细胞生长的影响.方法建立能够稳定表达、分泌NT-3蛋白的NIH3T3细胞系;选择出生2~4 d的BALA/CA小鼠,进行耳蜗螺旋神经节细胞原代培养并分为3组,其中Ⅰ组与稳定分泌NT-3的NIH3T3细胞共培养,Ⅱ组与传染空载体NIH3T3细胞共培养,Ⅲ组与NIH3T3细胞共培养,共培养2周后,观察各组螺旋神经节细胞的数量及轴突长度的变化.结果共培养2周后,与对照组相比,NIH3T3-NT-3共培养组SGC的数量以及突起的长度均显著高于对照组.结论NT-3可显著减轻耳蜗螺旋神经节神经的退行性改变.

Abstract:Objective: Co-culture the cochlear ganglion cells with this NIH3T3-NT3 cells, to observe the growth of SGCs in different condition. Methods: We estained the NIH3T3-NT3 cell line which stablely expresse NT-3 protein, then co-culture the SGCs with NIH3T3-NT3 cells, NIH3T3 cells transfected with pIRES2-EGFP and NIH3T3 cells respectively. After 2 weeks, the number of SGC and the length of axon in different groups were measured. Results: After co-culture 2 weeks, not only the number of the SGCs, but also the length of axon were more than the control groups. Conclusions: NT-3 could benefit the growth of SGCs in vitro.
