食管黏膜内微量元素含量与癌基因突变关系的研究

摘    要:目的 :探讨食管黏膜内微量元素含量、基因突变与食管病变的关系。方法 :15 1例食管活检标本分为正常、慢性炎、非典型增生与早期癌四种类型 ,以X 射线能谱仪扫描电镜进行微量元素定量检测 ,以S P免疫组化法检测PCNA表达及p5 3突变状态。结果 :Zn、Se、Mo随着食管上皮增生程度的升级含量渐减 ,不同组织学类型间差异有显著性 (P <0 .0 5 ) ,Cu Zn、Ni随着食管病变程度的升级含量渐增 ,差异有显著性 (P <0 .0 5 ) ,p5 3、PCNA阳性表达在正常上皮与非典型增生及早期癌组间差异有极显著性 (P <0 .0 1)。结论 :Zn、Se、Mo、Cu、Ni含量变化可能为高发区食管上皮p5 3突变与PCNA过表达的始动因素

Abstract:Objective To probe the relationship of trace element content and gene mutation in esophgeal lesions.Methods Esophageal biopsy specimen of 151 cases were divided into normal,chronic esophagitis,dysplasia and early carcinoma,and the quantitative determination of trace element was performed by using X ray energy spectrometry and the detection of PCNA expression and p53 mutation by using S P immunohistochemistry method.Results The content of Zn,Se,Mo was gradually reduced with upgrade of esophgeal epithelium hyperplasia.There was a significant difference in varied histological typies( P <0.05).The content of Cu/Zn,Ni was gradually increased in pace with upgrade of esophageal lesions,the difference significantce( P <0.05).The postiverates of p53 and PCNA were significant difference among normal esophageal epithelium,dysplasia and early carcinoma( P <0.01).Conclusion The content variation of Zn,Se,Mo,Cu,Ni could be the beginning factor for p53 mutation and PCNA overexpression of esophageal epithelium in high risk area.
