阿司匹林对结肠癌细胞株SW480生长增殖及NOS表达的影响

摘    要:目的观察阿司匹林对结肠癌细胞株SW480生长增殖、NOS表达及NO含量的影响并探讨其作用机制。方法以不同浓度(2.5、5.0、10.0 mmol/L)的阿司匹林干预体外培养人结肠癌细胞株SW480,采用生长曲线法和MTT法观察其对生长增殖的影响;免疫细胞化学染色法检测NOS的表达;Griess法测定培养基中NO含量。结果阿司匹林对结肠癌细胞株SW480生长增殖具有抑制作用,诱导iNOS的表达,增加NO的生成,并呈浓度依赖关系。结论阿司匹林可抑制结肠癌细胞株SW480的生长增殖;诱导iNOS的表达、增加NO含量可能是其部分机制。

Abstract:Objective To investigate the effect of Aspirin on the proliferation and NOS expression in colorectal cancer cell line SW480, and probe into its mechanism. Methods The effect of Aspirin on the growth of Colorectal Cancer cells was evaluated by MTT assay; NOS protein levels were determined by immunocytochemistry, NO concentration in the medium were determined by Griess assay. Results Aspirin inhibit the growth of SW480 cells,induce the expression of iNOS, increase the concentration of NO in the medium in a concentration dependent pattern. Conclusion Aspirin inhibit the growth of colorectal cancer cell line SW480. Up-regulated iNOS expression resulting a increase of NO concentration are ascribed to mechanism of anti-proliferation activity of aspirin.
