摘    要:目的 探讨60Coγ射线小剂量辐射损伤对小鼠血液microRNA的影响及意义。方法 SPF级C57BL/6J小鼠接受0.5Gy全身照射后,分别于照射后6h、24h进行外周血白细胞计数。同时应用Agilent microRNA生物芯片筛选小鼠血液中差异表达microRNA。结果 microRNA芯片筛选出辐射后6h小鼠中差异表达miRNA共11个,其中7个上调, 4个下调。照射24h小鼠中差异表达miRNA共32个, 其中13个上调, 19个下调。且与未照射组比较表达差异有统计学意义(P<0.05)。0.5Gy 照射后,6h和24h 外周血白细胞总数与对照组相比无统计学意义(P>0.05)。miRNA表达变化优于血液中白细胞的变化。结论 microRNA在血浆中差异表达与辐射损伤有关,其有望成为辐射损伤新的血液标志物。

Abstract:Objective To ingestigate the diferiential expression profiles of miRNA induced by low dose radiation in mouse peripheral blood and to explore their main function and significances. Methods After SPF C57BL/6J mice expose to 0.5Gy single whole body radiation,total number of peripheral WBC were measured at 6h and 24h. The changes of miRNA expression in peripheral blood were by using Agilent miRNA microarrays after radiation of 6h and 24h. Results After 6h with 0.5Gy Co γirradiation, miRNA microarray revealed that 11 miRNA were differentially expressed, in which 7 miRNA up-regulated, 4 miRNA down-regulated. After 24h with 0.5Gy Co γirradiation, there are 32 miRNA were differentially expressed, in which 13 miRNA up-regulated, 19 miRNA down-regulated.Compared with control group, the total nunber of peripheral WBC didn’t decreased obviously(P< 0.05). Conclusion miRNA were invoved in the regulation of low dose radiation damage, and miRNA expression sigtures derived from mouse blood maybe be use as new biomarkers for exposure to low dose radiation.
